Snake venom disintegrins update: insights about new findings

چکیده
Snake venom disintegrins are low molecular weight, non-enzymatic proteins rich in cysteine, present the of snakes from families Viperidae, Crotalidae, Atractaspididae, Elapidae, and Colubridae. This family originated through proteolytic processing metalloproteinases (SVMPs), which, turn, evolved a gene encoding an A Disintegrin And Metalloprotease (ADAM) molecule. Disintegrins have recognition motif for integrins their structure, allowing interaction with these transmembrane adhesion receptors preventing binding to extracellular matrix other cells. gives wide range biological functions, including inhibition platelet aggregation antitumor activity. As result, many studies been conducted attempt use natural compounds as basis developing therapies treatment various diseases. Furthermore, FDA has approved Tirofiban Eptifibatide antiplatelet compounds, they synthesized structure echistatin barbourin, respectively. In this review, we discuss some main functional structural characteristics class potential therapeutic use.

Snake Venom Disintegrins and Cell Migration
Cell migration is a key process for the defense of pluricellular organisms against pathogens, and it involves a set of surface receptors acting in an ordered fashion to contribute directionality to the movement. Among these receptors are the integrins, which connect the cell cytoskeleton to the extracellular matrix components, thus playing a central role in cell migration. Integrin clustering a...
متن کاملEvolution of snake venom disintegrins by positive Darwinian selection.
PII-disintegrins, cysteine-rich polypeptides broadly distributed in the venoms of geographically diverse species of vipers and rattlesnakes, antagonize the adhesive functions of beta(1) and beta(3) integrin receptors. PII-disintegrins evolved in Viperidae by neofunctionalization of disintegrin-like domains of duplicated PIII-snake venom hemorrhagic metalloproteinase (SVMP) genes recruited into ...
متن کاملSnake venom metalloproteinases and disintegrins: interactions with cells.
Metalloproteinases and disintegrins are important components of most viperid and crotalid venoms. Large metalloproteinases referred to as MDC enzymes are composed of an N-terminal Metalloproteinase domain, a Disintegrin-like domain and a Cys-rich C-terminus. In contrast, disintegrins are small non-enzymatic RGD-containing cysteine-rich polypeptides. However, the disintegrin region of MDC enzyme...
متن کاملSnake venom disintegrins: novel dimeric disintegrins and structural diversification by disulphide bond engineering.
We report the isolation and amino acid sequences of six novel dimeric disintegrins from the venoms of Vipera lebetina obtusa (VLO), V. berus (VB), V. ammodytes (VA), Echis ocellatus (EO) and Echis multisquamatus (EMS). Disintegrins VLO4, VB7, VA6 and EO4 displayed the RGD motif and inhibited the adhesion of K562 cells, expressing the integrin alpha5beta1 to immobilized fibronectin. A second gro...
متن کاملSnake Venom, Anti-snake Venom & Potential of Snake Venom
Many active secretions produced by animals have been employed in the development of new drugs to treat diseases such as hypertension and cancer. Snake venom toxins contributed significantly to the treatment of many medical conditions. Venomous snakes have a bad reputation and rightly so because of their often deadly bites. But what makes a snake’s bite so deadly is the venom. Of the 3000 snake ...
